Initial Assays
Incubate compounds with reagents or cells to begin screening. Incubation times vary: biochemical gPCR-based assays may require milliseconds to bind, while cancer phenotype assays may require days for sufficient incubation. Automate the process with a Beckman Coulter Life Sciences Biomek i-Series Automated Liquid Handler. If assays are miniaturized, acoustically dispense reagents with a Beckman Coulter Life Sciences Echo Acoustic Liquid Handler. Ensure incubation times are appropriate for screening methods and aims. Screening readouts will require different instruments for detection, such as plate readers, high content imagers, flow cytometers or even high throughput mass spectrometers. Readouts may include a fluorescent response, ATP assay, live/dead imaging of cells, mass spectra, etc. As readouts are recorded, automatically calculate plate-based QC metrics, analyze initial assay readouts and select top tips with Genedata's Screener software.
