Cell Growth
Picked colonies are inoculated into liquid media containing an antibiotic to select for bacterial cells expressing the plasmid and prevent contamination. Growth is controlled to precise optical density (OD) to maximize plasmid fermentation and prevent culture overgrowth and media starvation. Automate culture transfers on a Beckman Coulter Biomek i-Series Automated Liquid Handler and monitor OD on a Molecular Devices SpectraMax plate reader to ensure best conditions for plasmid harvest. Ensure full traceability of growth conditions using IDBS Polar Biopharma Lifecycle Management software.
