Functional Assessment of CRISPR-Edited Cells

CRISPR-edited cells require functional validation beyond genomic confirmation to demonstrate that genetic modifications result in the intended biological outcomes. Cells must be evaluated for potency, viability, toxicity and phenotype to allow researchers to assess whether CRISPR-mediated edits translate into measurable cellular responses relevant to the intended application. Readouts, including a fluorescent response, ATP assay, live/dead imaging of cells, or other image-measured responses, are performed with a Molecular Devices multi-mode plate reader with GxP-compliant software.

Cell-based assays are leveraged for functional assessments and require the ability for live-cell and time-lapse imaging. A Leica Microsystems STELLARIS Confocal Microscope Platform or Leica Microsystems Mica Microhub fulfills these requirements for imaging and cell-based functional assays.
