How AEMS takes your analysis to the new level

The Echo® MS+ system uses Acoustic Ejection Mass Spectrometry (AEMS). Samples are placed into compatible microtiter plates that are held in the Echo® MS+ system autosampler. Here, sound energy is applied to the bottom of each well individually. The sound energy causes reproducible droplets to be ejected from the well for capture in a carrier solvent of the OPI for dilution and transfer to the mass spectrometer source. From this point, the diluted sample is ionized using conventional electrospray ionization, making it ready for detection.

Accelerating Turnaround Time In High-Throughput ADME Pipelines

Accelerated Pharmacokinetic Profiling And Metabolite Monitoring Using Echo® Ms System
Here, the Echo MS System was used for early discovery phase studies providing DMPK data in seconds as opposed to the hours required when using traditional LC-MRM based scanning methodologies. The system provided highly quantitative data with excellent %CV and accuracy. The workflow also provided clearance information with good agreement between AEMS vs traditional LC-MS as well as high-throughput metabolite monitoring enabling accurate decisions on candidate progression.

Ultra-Fast, Accurate And Simultaneous Quantification Of Ritonavir And Lopinavir In Human Plasma
Lopinavir and ritonavir are two protease inhibitors (class of anti-viral drugs) often used in a fixed-dose combination, along with other medications, for prevention and treatment of HIV/AIDS. Monitoring of lopinavir and ritonavir in human plasma is needed to ensure proper therapeutic levels are maintained. A very high throughput bioanalytical method (3 sec/sample) for the simultaneous analysis of ritonavir and lopinavir in human plasma is described using the Echo MS platform. Using a simple liquid-liquid extraction (LLE) for sample preparation, the assay provided very sensitive, accurate and reproducible results.

Up To 10X Increased Throughput For Metabolic Stability Assays
ADME profiling is typically performed on candidates later in the development cycle but performing these screens earlier would provide significant benefits. However, significant gains in sample analysis throughput would be required as the number of compounds to be tested would likely increase by an order of magnitude. Acoustic Ejection Mass Spectrometry (AEMS) on the Echo® MS system is a very high throughput quantitative technique that is ideal for labs performing in vitro ADME. Here using a metabolic stability assay, a ~10x increase in sample throughput was demonstrated when using the Echo® MS system coupled with LeadScape Analyze software as compared to an LC-MS/MS approach.
